The Division of Molecular Genetics at Columbia University Irving Medical Center in New York City is seeking a highly motivated Associate Research Scientist to immediately participate in the conduct of studies on the molecular and cellular mechanisms of Prader-Willi syndrome, using stem cell models of human disease. The applicant will be using various techniques including the genetic modification of stem cells, directed differentiation, flow cytometry, immunocytochemistry, single molecule analysis of replicating DNA, and whole genome copy number analysis. KEY RESPONSIBILITIES The candidate will focus on genome stability and precise gene editing in early human embryos. The candidate will be working on: "Correcting Mutations in Blastocysts in the Mammalian Preimplantation Embryo". and: "checkpoint activity in mammalian preimplantation development". The responsibilities include embryology, embryo manipulation, whole genome amplification, production of mRNAs for DNA repair factors, transcriptional and proteomic analysis of mammalian embryos across species, laboratory maintenance (lab jobs), genetic analysis, manuscript writing, mentoring of junior students, postdocs and technicians. Techniques involve epigenetic reprogramming into iPSCs, somatic cell nuclear transfer, CRISPR-based gene editing approaches, chromosomal analysis through next generation sequencing, and immunostaining.
